A personalized mRNA melanoma cancer shot developed by Moderna and Merck using an AI generated design has met the primary endpoint in a late-stage phase 3 clinical trial.1 In August, the two pharmaceutical giants reported that, when combined with an existing treatment, the vaccine reduced the risk of melanoma returning or spreading.
Cancer experts are enthusiastic about the possibility of a cancer treatment, known as intismeran autogene, calling it the beginning of an era of “custom-made vaccines” that train a person’s immune system to attack cancer cells by targeting mutations unique to the individual patient’s tumors.2 Moderna prefers not to use the term “vaccine” to describe its biological product, but rather calls it an individualized neoantigen therapy.
“It’s an algorithm as much as it is therapy,” said Jane Healy, MD, PhD, Merck’s head of oncology development.3
“I think this is really an important achievement,” said Jedd Wolchok, MD, PhD, director of the Meyer Cancer Center at Weill Cornell Medicine. “It tells us what might have been considered science fiction 15 or 20 years ago – that we can produce a personalized vaccine based on sequencing from a tumor from a pathology specimen – could be used to delay progression of melanoma.3
Merck and Moderna have been building the tailored shot using machine learning algorithms for more than 10 years. While “neoantigen” shots have long been seen as a potential cancer treatment, Merck and Moderna’s trials were the first to show “clinically meaningful” results.3
mRNA Cancer Shot to Be Paired With Existing Cancer Drug Keytruda
Researchers tested the melanoma cancer shot on patients with early-stage melanoma following surgical removal of tumors. They paired the vaccine with Merck’s drug Keytruda, which is currently approved and used in 20 different kinds of cancerous tumors and 45 types and stages of cancer. Keytruda activates the body’s immune system T-cells that seek out abnormal cells.4
“The whole special thing about Keytruda was that it would unlock the dogs of war,” Dean Li, MD, PhD, president of Merck Research Laboratories, said. The vaccine provides 34 patient-specific cancer targets for those dogs to recognize, he said, and only one needs to work. “With the personalized cancer treatment, the dog has your scent.”2
mRNA Cancer Shot Would Use Computer to Determine Genetic Instructions
If approved, intismeran autogene would be the first medicine whose genetic instructions are determined by a computer algorithm using AI technology. The shot is personalized by taking a sample of a patient’s tumor and using computer analysis to identify potentially tens of thousands of unique mutations. Researchers then select specific mutant proteins to encode in the shot. Once administered, the patient’s cells produce those proteins, prompting the immune system to recognize and mount an immune response.4
Moderna President Steven Hoge noted that the companies hope to file with the Food and Drug Administration as quickly as possible, citing months not years.3

Take a minute and read this again very very carefully. If one’s bodies did not mount an immune response to the deformed proteins in the first place, what makes you think that encoding your cells to produce more of these will help? Are they encoding the mutation into the genome itself?
Know what else also works; peptides, ivermectin, concentrated cbd oils, rife wave frequency generators.
